The earlier report in Famitsu magazine has been officially confirmed by Capcom. It's not just co-op, either. The new game to feature usage of the Zapper peripheral on the Wii will also include:
Locations from previous games (Resident Evil 0, 1, 2 and 3) in addition to new ones, and will focus on the series' backstory surrounding the fall of the Umbrella Corporation.
It makes sense geographically that there won't be any locations from RE4, but whether or not any enemies from that game will make it into Umbrella Chronicles isn't clear yet. Anyway, two-player zombie killing a la House of the Dead, but set in the Resident Evil universe? This gets my full endorsement.
